PT-141 initiates cellular signaling by binding to specific subtypes within the melanocortin receptor family. These receptors belong to the G protein-coupled receptor superfamily, and activation raises intracellular cyclic adenosine monophosphate levels. This cascade ultimately influences neuronal circuits in the central nervous system that are associated with sexual desire and arousal. Research indicates the compound's action concentrates in hypothalamic regions rather than peripheral tissues, which helps explain some observed pharmacological features. The selectivity of receptor binding underlies its functional differences.
Compared with the related compound melanotan II, PT-141 shows markedly weaker activation of receptors tied to pigmentation. This difference stems from subtle structural variations that alter affinity distribution across receptor subtypes. Investigators propose that such selectivity produces a different side effect profile in specific applications. However, downstream consequences of prolonged receptor activation remain uncertain in the literature. Published studies do not fully agree on the duration of signaling pathway activity and the mechanisms of desensitization.
From a pharmacokinetic standpoint, the peptide is usually delivered by injection because oral bioavailability is very low; proteases in the digestive tract degrade it rapidly. After subcutaneous administration, plasma concentrations reach a peak within roughly one hour. Its elimination half-life is relatively short, with most reports placing it in the range of a few hours. Nasal formulations have also been examined, though absorption varies widely between individuals. Metabolism proceeds mainly through peptidase cleavage, and the resulting products are excreted by the kidneys.
PT-141 is the research designation for bremelanotide, a synthetic cyclic heptapeptide developed as a melanocortin receptor agonist. The peptide contains seven amino acids with an internal lactam bridge that constrains the backbone into a stable ring. Cyclisation of this kind improves resistance to enzymatic degradation relative to linear analogues. The compound was first explored for effects on melanin production, because several melanocortin receptors influence pigmentation. Its later association with sexual desire emerged from observations made during that early work, and the molecule became the subject of a separate development programme.
Bremelanotide is a moderately large peptide with a molecular mass near 1025 daltons. In lyophilised form it appears as a white to off-white powder and is freely soluble in water and other polar solvents. The intact lactam ring is essential for receptor affinity, while linearised fragments bind far more weakly. Solutions are sensitive to extremes of pH and to prolonged exposure to light and heat, so handling typically involves buffered conditions and cold storage. Its short plasma half-life reflects rapid distribution and clearance rather than chemical breakdown inside the vial.
Development of the peptide passed through several delivery formats, including an intranasal version tested in early trials and an injectable version that entered later clinical study. Regulatory approval for a subcutaneous product in the United States was granted in 2019 after review of controlled trials in premenopausal women. Outside the clinic, the compound circulates in research and non-pharmaceutical markets under its code name, where identity and purity vary considerably between suppliers. Synonyms appearing across technical literature include bremelanotide, PT-141, and Palatin 141.

Reconstitution is usually performed with sterile water or a dilute acetic acid solution, and the choice of solvent affects both dissolution speed and final pH. Complete dissolution should be confirmed by visual inspection before any aliquot is taken, since undissolved particles can concentrate in the sampling volume. Repeated freeze-thaw cycles are the most common cause of gradual loss of purity, so dividing a stock into single-use aliquots at the first opportunity is standard practice. Working solutions kept refrigerated are generally used within days rather than weeks.
Identity and purity are established with orthogonal methods rather than a single test. Reverse-phase high-performance liquid chromatography with ultraviolet detection gives a purity figure by area normalization, while mass spectrometry confirms the expected molecular ion. Amino acid analysis or peptide mapping can detect sequence errors that a mass value alone would miss, and residual counterion content is sometimes measured separately. Common impurities include truncated sequences, oxidized residues, and deamidated products; reporting them individually is more informative than a single composite purity number.
Bremelanotide functions as an agonist at several melanocortin receptor subtypes, with the strongest functional activity reported at the MC4 subtype. MC4 receptors sit in hypothalamic circuits that influence appetite, energy balance, and components of sexual behaviour. Rodents lacking functional MC4 receptors show altered mating behaviour, which supports a role for this pathway in desire. The precise sequence of events connecting receptor activation to reported human effects remains only partly characterised. Because the same receptor family governs pigmentation and inflammatory signalling, selectivity is a recurring theme in pharmacological discussion.
Clinical programmes in this area have relied mainly on randomised, double-blind, placebo-controlled designs in premenopausal women. Primary endpoints usually combine a validated questionnaire covering desire domains with counts of satisfying sexual events and a separate measure of distress. Reported outcomes show statistically significant but modest average improvement over placebo, with wide individual variation. Adverse events such as nausea, flushing, and headache occur frequently and can limit tolerability. Whether short-term trial gains translate into lasting change for most users is an open question.
Evidence outside the studied population is sparse. Trials have concentrated on premenopausal women with a defined diagnosis, and data for postmenopausal women, men, and people taking interacting medications remain limited. Non-prescription use of the peptide for comparable goals is widespread but is not supported by published controlled data. Observed changes in blood pressure have drawn attention to cardiovascular monitoring during use. The literature generally frames the compound as a targeted receptor agonist rather than a general libido enhancer, and basic questions about mechanism and long-term safety are unresolved.
For SNP annotation, many kinds of genetic and genomic information are used. Based on the different features used by each annotation tool, SNP annotation methods may be split roughly into the following categories: Genomic information from surrounding genomic elements is among the most useful information for interpreting the biological function of an observed variant. Information from a known gene is used as a reference to indicate whether the observed variant resides in or near a gene and if it has the potential to disrupt the protein sequence and its function. Gene based annotation is based on the fact that non-synonymous mutations can alter the protein sequence and that splice site mutation may disrupt the transcript splicing pattern.
This type of specificity is sensitive to the substrate's optical activity of orientation. Stereochemical molecules differ in the way in which they rotate plane polarized light, or orientations of linkages (see alpha, beta glycosidic linkages). Enzymes that are stereochemically specific will bind substrates with these particular properties. For example, beta-glycosidase will only react with beta-glycosidic bonds which are present in cellulose, but not present in starch and glycogen, which contain alpha-glycosidic linkages. This is relevant in how mammals are able to digest food. For instance, the enzyme Amylase is present in mammal saliva, that is stereo-specific for alpha-linkages, this is why mammals are able to efficiently use starch and glycogen as forms of energy, but not cellulose (because it is a beta-linkage). Specific equilibrium dissociation constant for formation of the enzyme-substrate complex is known as k d {\displaystyle k_{d}}
AlphaKnot is a scientific database and web server for detecting, classifying, and visualizing protein knots and other forms of protein-chain entanglement. It was developed to facilitate the analysis of protein structures predicted by AlphaFold and other machine-learning methods, but can also be used to analyze experimentally determined structures. The current version, AlphaKnot 2.0, combines two closely related components: a precomputed database containing proteins identified as knotted in large-scale structure-prediction datasets, and an analysis server that allows users to investigate the topology of individual protein structures in greater detail.

AREs are recognized by RNA binding proteins such as tristetraprolin (TTP), AUF1, and Hu Antigen R (HuR). RNA-binding proteins that bind AREs have been termed ARE-BPs and as of 2019, about 20 ARE-BPs have been identified. Although the exact mechanism is not very well understood, recent publications have attempted to propose the action of some of these ARE-BPs. One characteristic of many ARE-BPs is that they can shuttle between the nucleus and cytoplasm and exert different functions to control gene expression based on their subcellular location. AUF1, also known as hnRNP D, binds AREs through RNA recognition motifs (RRMs). AUF1 is also known to interact with the translation initiation factor eIF4G and with poly(A)-binding protein, indicating that AUF1 senses the translational status of mRNA and decays accordingly through the excision of the poly(A) tail.
81. ArXiv [Preprint]. 2026 Jul 29:arXiv:2605.17186v2. Operator splitting for exploiting linear-rate closure in solving infinite ODE hierarchies. Chang JC. We introduce an operator-splitting method for infinite hierarchies of linear ordinary differential equations (ODEs) indexed by nonnegative integers. When the coupling coefficients depend linearly on the count index, an exact transformation closes the equations on finite count-index windows without an upper-boundary value. For more general hierarchies, Strang splitting applies the linear-rate closure during the linear-rate substeps and a conventional capped solver to the remainder. We derive the closure from generating functions and the method of characteristics and extend it to multi-indexed systems. The derivation requires neither positivity nor mass conservation, so it applies to a wider class of systems than the examplar stochastic models presented here. We discuss branching processes, stochastic predator-prey dynamics, the Schlögl chemical kinetics model, and a telegraph model for gene expression. Through numerical experiments and computational cost analyses we demonstrate that our operator splitting method is typically advantageous for solving large scale systems in terms of memory usage and computational time, while retaining accuracy competitive with finite state projection (FSP) methods. After replication of the desired region, the RNA primer is removed by DNA polymerase I via the process of nick translation. The removal of the RNA primer allows DNA ligase to ligate the DNA-DNA nick between the new fragment and the previous strand. It primarily activates specific subtypes in the melanocortin receptor family. These receptors are G protein-coupled and mediate signaling mainly within the central nervous system.
It activates pigmentation-related receptors more weakly. This selectivity is thought to alter its side effect profile.
Injection is the most common route. Nasal administration has been studied as well, though absorption varies considerably.
PT-141 is a laboratory code for bremelanotide, a synthetic peptide that activates melanocortin receptors. It has been developed both as an injectable pharmaceutical product and as a research chemical. The two contexts differ sharply in how identity, purity, and dosing are controlled.
